- Toll-like receptor 1/2 ligand enhances IL-4 receptor signaling in human Langerhans cells., International Investigative Dermatology 2018, 2018/05, Without Invitation, English, Toll-like receptor 2 (TLR2) on human dendritic cells (DCs) recognizes bacterial components from Staphylococcus aureus which is present in over 80 % of atopic dermatitis (AD) skin lesion. DCs secrete cytokines and chemokines, and thus orchestrate T cell responses. In AD, it is considered that Th2 inflammatory response dominate the acute phase. IL-4 plays an important role in AD patients and treatment with Dupilumab, a monoclonal antibody against IL-4 receptor subunit alpha (IL-4R), shows significantly improvements for AD patients. TLR2 is expected to have an interaction with IL-4R. Thus, we investigated the functional relevance between TLR2 and IL-4R in human Langerhans cells (LCs).
LCs (CD34LCs) were generated from CD34+ progenitor cells in vitro. CD34LCs were treated with IL-4 and/or TLR1/2 agonist Pam3Cys and IL-4R complexes were analyzed on mRNA and protein level by real-time PCR (RT-qPCR) and flow cytometry (FCM), respectively. The effects of Pam3Cys pretreatment on IL-4R signaling via signal transducer and activator of transcription 6 (STAT6) phosphorylation analysis and production of Th2 chemokines in CD34LCs were investigated by FCM, RT-qPCR and enzyme-linked immunosorbent assay.
Although mRNA levels of all IL-4R subunits were increased with Pam3Cys stimulation, only IL-2 receptor subunit gamma (IL-2R) was upregulated on CD34LCs cell surface. STAT6 was activated by IL-4 and even stronger by pretreatment with Pam3Cys. Pam3Cys alone did not activate STAT6. CCL17 synergistically increased with combined stimulation, while CCL13 increased additively. Anti-IL-4R antibody inhibited the production of CCL17 and CCL13 by IL-4.
These results show that Pam3Cys enhances IL-4R signaling by upregulating the expression of IL-2R Hence, the inflammatory response induced by IL-4 is more likely to be caused in patients with AD who are susceptible to S. aureus colonization.
